Hyperglycemic clamp to assess insulin secretion and β-cell function in conscious mice
Following overnight fast, a 2-hr hyperglycemic clamp will be conducted in awake mice with a variable infusion of 20% glucose to raise and maintain plasma glucose concentration at ~300 mg/dl. Blood samples will be collected at 10~15 min intervals for the measurement of plasma glucose and insulin concentrations. The area-under-curve of plasma insulin levels and glucose infusion rate to maintain hyperglycemia will reflect pancreatic β-cell function in vivo.
